    The CHAIN-REDS Semantic Search Engine

    e-Infrastructures, and in particular Data Repositories and Open Access Data Infrastructures, are essential platforms for e-Science and e-Research and are being built since several years both in Europe and the rest of the world to support diverse multi/inter-disciplinary Virtual Research Communities. So far, however, it is difficult for scientists to correlate papers to datasets used to produce them and to discover data and documents in an easy way. In this paper, the CHAINREDS project’s Knowledge Base and its Semantic Search Engine are presented, which attempt to address those drawbacks and contribute to the reproducibility of science

    Different patterns of cortical excitability in major depression and vascular depression: a transcranial magnetic stimulation study

    BACKGROUND: Clinical and functional studies consider major depression (MD) and vascular depression (VD) as different neurobiological processes. Hypoexcitability of the left frontal cortex to transcranial magnetic stimulation (TMS) is frequently reported in MD, whereas little is known about the effects of TMS in VD. Thus, we aimed to assess and compare motor cortex excitability in patients with VD and MD. METHODS: Eleven VD patients, 11 recurrent drug-resistant MD patients, and 11 healthy controls underwent clinical, neuropsychological and neuroimaging evaluations in addition to bilateral resting motor threshold, cortical silent period, and paired-pulse TMS curves of intracortical excitability. All patients continued on psychotropic drugs, which were unchanged throughout the study. RESULTS: Scores on one of the tests evaluating frontal lobe abilities (Stroop Color-Word interference test) were worse in patients compared with controls. The resting motor threshold in patients with MD was significantly higher in the left hemisphere compared with the right (p < 0.05), and compared with the VD patients and controls. The cortical silent period was bilaterally prolonged in MD patients compared with VD patients and controls, with a statistically significant difference in the left hemisphere (p < 0.01). No differences were observed in the paired-pulse curves between patients and controls. CONCLUSIONS: This study showed distinctive patterns of motor cortex excitability between late-onset depression with subcortical vascular disease and early-onset recurrent drug resistant MD. The data provide a TMS model of the different processes underlying VD and MD. Additionally, our results support the “Vascular depression hypothesis” at the neurophysiological level, and confirm the inter-hemispheric asymmetry to TMS in patients with MD. We were unable to support previous findings of impaired intracortical inhibitory mechanisms to TMS in patients with MD, although a drug-induced effect on our results cannot be excluded. This study may aid the understanding of the pathogenetic differences underlying the clinical spectrum of depressive disorders

    CHAIN-REDS DART Challenge

    CHAIN-REDS (Coordination and Harmonisation of Advanced e-infrastructure for Research and Education Data Sharing) is EU project focused on promoting and supporting technological and scientific collaboration across different communities established in various continents. Nowadays, one of the most challenging scenarios scientist and scientific communities are facing is huge amount of data emerging from vast networks of sensors and form computational simulations performed in a diversity of computing architectures and e-infrastructure. The new knowledge coming out from the interpretation of these datasets, reported on the scholar literature, is increasingly problematic to be reproducible due to the difficulty to access measured data repositories and/or computational applications that generate synthetic data through computer simulations. This paper presents CHAIN REDS approach, several tools and services, based on the adoption of standards, aimed at providing easy/seamless access to datasets, data repositories, open access document repositories and to the applications that could make use of them. All these tools and services are enclosed in what we have called the Data Accessibility, Reproducibility and Trustworthiness (DART) challenge. This initiative allows researchers to easily find data of his interest and directly use them in a code running by means of a Science Gateway (SG) that provides access to cluster, Grid and Cloud infrastructure worldwide. In this scenario, the datasets are found by means of either the CHAIN-REDS Knowledge Base (KB) or the Semantic Search Engine (SSE), the applications ran on the CHAIN-REDS SG, accessible through an Identity Federation. The datasets can be both identified by Persistent Identifier (PID) and assigned unique number ID. Scientists can then access the data and the corresponding application in order to either reproduce and extend the results of a given study or start a new investigation. The new data (and the new paper if any) are stored on the Data Infrastructure and can be easily found by the people belonging to the same domain making possible to start the cycle again.Repositório de dados científicos.Ibero-American Science and Technology Education Consortium (ISTEC

    Critical role of maternal selenium nutrition in neurodevelopment: Effects on offspring behavior and neuroinflammatory profile

    Research in both animals and humans shows that some nutrients are important in pregnancy and during the first years of life to support brain and cognitive development. Our aim was to evaluate the role of selenium (Se) in supporting brain and behavioral plasticity and maturation. Pregnant and lactating female rats and their offspring up to postnatal day 40 were fed isocaloric diets differing in Se content - i.e., Optimal, SubOptimal, and Deficient - and neurodevelopmental, neu-roinflammatory, and anti-oxidant markers were analyzed. We observed early adverse behavioral changes in juvenile rats only in SubOptimal offspring. In addition, SubOptimal, more than Deficient supply reduced basal glial reactivity in sex dimorphic and brain-area specific fashion. In female offspring, Deficient and SubOptimal diets reduced the antioxidant Glutathione peroxidase (GPx) activity in the cortex and in the liver, the latter being the key organ regulating Se metabo-lism and homeostasis. The finding that Se SubOptimal was more detrimental than Se Deficient diet may suggest that maternal Se Deficient diet, leading to a lower Se supply at earlier stages of fetal development, stimulated homeostatic mechanisms in the offspring that were not initiated by SubOptimal Se. Our observations demonstrate that even moderate Se deficiency during early life negatively may affect, in a sex-specific manner, optimal brain development

    New genetic loci link adipose and insulin biology to body fat distribution.

    Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms
